The BABL/C mice were immunized with the compound of Trx-LHRH and trx antibody. When the titre came to 1:104, the spleen cells were fused with the SP2/0 by PEG-1500. The hybridoma surprunt was detected with indirect ELISA and inhibitory ELISA. The hybridoma strains D4B7 and B6E6 secreting Mcab against LHRH were obtained by dilluting several times and the ascite was identified with Westen-blot. A double antibody sandwich ELISA was established to detect the quantity of effective content of the emasculating vaccine.RT-PCR was established from the hypothalamus of rats, whose ages were l-2days,7-14days and 6-8weeks ,to analysize the expression of LHRH mRNA level ,the results showed that rats of l-2clays and 6-8weeks could be detected of LHRH and p-Actin lines .while 7-14 days only could be detected of β-Actin. It suggested that LHRH mRNA began to decrease when the rats was born 1 -2 days and recommended to express activating sexual development. The results coincided with the changes of LHRH content, Which made a basis for investigating the effect of LHRH in sex maturation.The immature and mature mice were immunized with the emasculating vaccine. The mating ability, the proportion of pregnancy, the morphological and histological changes of testicles, the proportion of active sperms of mice and testosterone were examined . The results showed that the immunized mice could recover after a interval time .The SD rats were immunized with emasculating vaccine with adjuvants of alums, A1(OH)3 and FA. The titer of LHRH antibody, testosterone level, the morphological and histological changes of testicles and prostates the proportion and number of freaky sperms were compared among the three groups .The results showed the three kinds of emasculating vaccines could make the rats gain weight and decreasetestosterone. As FA vaccine made a great influence on antibody and testosterone and the alum vaccine had a better effect on regressing prostates.
