This study use F8 RIL population of Opata5 W7984 and nulli-tetrasome based on Chinese Spring as material, through analysis express sequence tags of wheat and its near species-genus, we constructed genetic linkage maps with designed EST-SSR. At the same time, we also select tightly linkaged SSR markers with powdery mildew resistance genes Pm2 and Pm21 in wheat by using three NIL materials, Bainong3217 Mardler, Beijing837 Mardler and Bainong3217 Pm21 and one F2 population of Bainong3217 Pm21 respectively, the results as following:1. Analysis EST sequence download from EST database, and designed 597 pairs of SSR primers with Primer3, DNASTAR and OLIGO etc, and also we gather primers' characteristics, such as annealing temperature, amplified segment length, quantity of amplify, background of the primer amplify etc after PCR and polyacrylamide gelatinous electrophoresis.2. Use Opata85 and W7984 as parental materials, selected 109 out of 597 pairs of SSR primers who showed polymorphism. And also, we use F8 RIL population of Opata5 W7984 do population test, and finally we mapped 65 loci on wheat genetic linkage maps finely.3. Use 84 Pairs of SSR markers (Xgwm serials) test 36 nulli-tetrasome based on Chinese Spring, we certificated 16 ture nulli-tetrasome materials, we use 4 of them located 53 pairs of primers on wheat chromosomes respectively. The other 20 nulli-tetrasome materials are not ture.4. Use nearly isogenic lines Mardler X Bainong3217 and Mardler X Beijing837, selected SSR primers of wheat 5DS chromosome to find tightly linkaged markers with Pm2 gene resist to wheat powdery mildew, but found no polymorphism between resistant material and infectional material. Subsequently, we converted RFLP marker BCD1871 which tightly linkaged toPm2 gene and EST sequence (BE404135) on the end of 5DS chromosome into three pairs of STS marekrs and three pairs of SSR markers respectively, but found no polymorphism between resistant material and infectional material also.5. Use nearly isogenic lines Pm21 Bainong3217, selected SSR primers of wheat 6AS chromosome to find tightly linkaged markers with powdery mildew resistance gene Pm21 in wheat, but found that all these primers only amplified in infectional materials.
