Ganoderma lucidum is a famous medicinal fungus .Its reproductive cell, spores, contain abundant physiological activity substances, such as polysaceharides, triterpenes, peptides etc., which had functions of anti-cancers, immunomodulation to human beings. In order to increase their contains absorbing rate, the spores, being hard and tough, had to be broken completely. Two physical spore-breaking methods, supersonic airflow and two-roller spindle, were developed and compared with Homo-mixing method. Two-roller spindle was the best method for breaking spores of G. lucidum. The average rate was 97.4%. Compared with intact ones, sporoderm-broken spores reducing carbohydrates in water and in acid solution were increased, added up to 19%, 11.8% respectively; dissolved substances were different by UV absorption; appearance characteristics were also different; And their physiological functions~ DTH and DC5O, were more notable. Sporoderm-broken spores were also able to improve immunoreaction. Primary study was experienced to control the oxidation of sporoderm- broken spores by different antioxidants, such as tea polyphenol (TPP), TBHQ, ascorbic acid (Vc). The best composition was with 200ppm Vc and lOOppm TPP. And sporodcrm-broken spores were also safety by the experiments based on GB15193-94.
