Radix Ophiopogonis was the root of Liliaceae Ophiopogonis japonicas(L. f.)Ker-Gawl.A large number of literature reports indicae that Radix Ophiopogonis have many pharmacological activities in it, Such as anti-arrhythmic, anti-carcinogenic, anti-inflammatory, anti-shock and anti-aging. Sichuan and Zhejiang is the traditional origin of Radix Ophiopogonis. Radix Ophiopogonis produced in Zhejiang is known as Hang Radix Ophiopogonis, wich is cultivated in the city of Cixi, Yuyao, Xiaoshan, and it is traditional Daodi Chinese medicinal materials. Radix Ophiopogonis produced in Sichuan is called Chuan Radix Ophiopogonis, wich is is mainly produced in Mianyang, Santai. In this study, to provide theoretical basis for elucidating the mechanism of Radix Ophiopogonis Daodi Chinese medicinal materials, we studyed the growth environment, cultivation methods, genetic background, effective composition, pharmacological activities of Hang Radix Ophiopogonis and Chuan Radix Ophiopogonis and mainly achieved the following results.1The different cultivation methods between Hang Radix Ophiopogonis and Chuan Radix Ophiopogonis is that Hang Radix Ophiopogonis is cultivated for three years and Chuan Radix Ophiopogonis grows for one year. The content of total N and available phosphorus in Hang Radix Ophiopogonis cultivated soil was significantly lower than that in Chuan Radix Ophiopogonis, PH value is significantly higher than Chuan Radix Ophiopogonis.2Using EST-SSR markers that were established based on Radix Ophiopogonis transcriptome data to determine the genetic diversity of Radix and its related species. The results show that there is a clear genetic difference between Hang Radix Ophiopogonis and Chuan Radix Ophiopogonis.3The contents of Methylophiopogonanone A, Methylophiopogonanone B, ophiopogonin B, and ophiopogonin D’in Hang Radix Ophiopogonis are higher than Chuan Radix Ophiopogonis.4AUPLC/Q-TOF-MS based metabonomic method was developed for quality evalution of Hang Radix Ophiopogonis and Chuan Radix Ophiopogonis. Identified15kinds of steroidal saponins, the contents of ophiopogonin H、ophiopojaponin R and ophiofurospiside F in Hang Radix Ophiopogonis are higher than Chuan Radix Ophiopogonis, and the contents of ophiopogonins H、(2a,3p,22R,25S)-3-{[p-D-Glucopyranosyl-(1->2)-β-D-glucopyranosyl-(1->2)-β-D-gl ucopyranosyl]oxy}-2,22-dihydroxyfurost-5-en-26-ylβ-D-glucopyranoside in Hang Radix Ophiopogonis are lower than Chuan Radix Ophiopogonis. PCA analysis showed that the chemical compositions in water extract are obviously different between Hang Radix Ophiopogonis and Chuan Radix Ophiopogonis, biennial Hang Radix Ophiopogonis and three years Hang Radix Ophiopogonis,2012Chuan Radix Ophiopogonis and2013Chuan Radix Ophiopogonis.5After healthy rats were gavaged with Radix Ophiopogonis, Hang Radix Ophiopogonis and Chuan Radix Ophiopogonis both can reduce the contents of LD and LDH in myocardial, increased GSH-PX content in blood, but there are some differences between Hang Radix Ophiopogonis and Chuan Radix Ophiopogonis.C low dose group rats myocardial’s extent of LD was significantly lower than Hang Radix Ophiopogonis low dose group. High dose Chuan Radix Ophiopogonis elevated GSH-PX level in serum significantly lower than the high dose ang Radix Ophiopogonis. In addition five Radix Ophiopogonis administration groups total antioxidant capacity increased in rat lung.6After healthy rats were gavaged with Radix Ophiopogonis, rat organs RNA-seq analysis results showed that10differentially expressed genes were found in rat lungs after gavaged Hang Radix Ophiopogonis,4genes were significantly upregulated and6genes were downregulated,7genes are directly or indirectly involved in the lungs immunomodulatory or lung tissue composition. In comparison with the control group no differentially expressed gene was found in the group gavaged with Chuan Radix Ophiopogonis. Hang Radix Ophiopogonis increased the expression of RETSAT and TRIM50in myocardial cells, thereby it can improve myocardial cell immunity and disease resistance. Chuan Radix Ophiopogonis regulated the expression of IRX2、 IRX1、ANKRD23、CXCL11、EPHA3and so on, and they are related to myocardial cell differentiation, inhibition of hypoxia-inducible factor, inflammatory. Visiblly, Radix Ophiopogonis can partly improve the function of heart and lung, there are some differences in pharmacodynamic between Hang Radix Ophiopogonis and Chuan Radix Ophiopogonis.
