In this study,polysaccharide production for the domestic nebrodensis widespread adoption of fruiting body extract productivity due to resource constraints was low,and training through the Submerged Culture Mycelia of the low rate of extraction of polysaccharides,the economic benefits of the status was not significant, through submerged fermentation culture medium extract of Chinese herbal medicines added to increase the nebrodensis fermentation yield of polysaccharides. Fermentation process by measuring biomass,nebrodensis intracellular polysaccharide,exopolysaccharide,pH,density of mycelia ball,the ball diameter and mycelial fermentation broth viscosity fermentation indicators,such as a series of dynamic changes to the promotion of screening nebrodensis polysaccharide accumulated additive type and amount of medicine and studied the different methods of nebrodensis broken intracellular yield of polysaccharide.The study aimed at the development and utilization of polysaccharide nebrodensis and related enterprises to provide a theoretical basis.The results obtained as follows:1.Five kinds of Chinese herbal medicine on the growth of Pleurotus nebrodensis screening test:Plate culture method using five kinds of Chinese herbal derived growth promoting nebrodensis.The concentration of Radix Angelicae Sinensis 25mg/mL, 50mg/mL,75mg/mL,colony diameter 6.0cm,6.3cm,5.9cm respectively;The concentration of Radix Codonopsis Pilosulae 100mg/mL,125mg/mL,150mg/mL, colony diameter 6.2cm,6.5cm,6.0cm;The concentration of Astragalus membranaceus Bunge 5mg/mL,10 mg/mL,15mg/mL,colony diameter 5.2cm,5.5cm,5.8cm;The concentration of Glycyrriza Uralensis Fisch.G.Glabra L. 5mg/mL,10mg/mL,15mg/mL,colony diameter 5.6cm,5.8cm,6.0cm;The low concentrations of Radix Isatidis on the growth of nebrodensis did not has significant different,however high concentrations inhibited the growth of nebrodensis obviously.2.Test of effects of polysaccharide contents in which adding herbs to nebrodensis fermentation.The shake-flask culture through Submerged fermentation at three concentrations of four kinds of Chinese herbal,the results showed that the biomass, number of intracellular sugar had been prompted by herbs.Radix Angelicae Sinensis 50mg/mL,Radix Codonopsis Pilosulae 125mg/mL,Astragalus membranaceus Bunge 15mg/mL,Glycyrriza Uralensis Fisch.G.Glabra L 15mg/mL has the best results compared with control group,which biomass reached 1.46g/100mL, 1.40g/100mL,1.28g/100mL,1.31g/100mL;intracellular polysaccharide content respectively reached 10.58%,10.43%,8.80%,9.0%;Then the results also indicated that fermentation of nebrodensis extracellular multi-sugar content aslo had been prompted by herbs,Radix Angelicae Sinensis 75mg/mL,Radix Codonopsis Pilosulae 100mg/mL,Astragalus membranaceus Bunge 15mg/mL,Glycyrriza Uralensis Fisch.G.Glabra L 10mg/mL were the best results compared with control group,which extracellular polysaccharide content reached 8.03%,7.93%,5.58%, 5.66%.However intracellular polysaccharide of Radix Angelicae Sinensis,Radix Codonopsis Pilosulae had great effect than the extracellular polysaccharide,then the concentration of polysaccharide and Intracellular of Glycyrriza Uralensis Fisch.G.Glabra L.,Radix Isatidis did not has significant different comparing with each other.3.Methods of three broken nebrodensis intracellular yield of polysaccharideOrdinary mechanical grinding,ultrasound,Snailase method were used to broken cells,the best result was ultrasound broken method measureing with yield of nebrodensis.Form the experiment the nebrodensis yield as the following order: ultrasound broken method8.73%,snail enzyme7.53%,mechanical grinding6.3%.4.Determination to intracellular polysaccharide components of Pleurotus nebrodensis.The main compounds of 50mg/mL Radix had been identified with GC,the result showed the glucose32%,galactose39%,mannose23%respectively.
