Study On Extraction, Separation And Purification Of Alkaloids From Sophra Alopecuroides L.

Sophora alopecuroides L.a leguminous Plant of SoPhora,is widely distributed in the northwestern region of China.The extraction industry of Sophora alopecuroides L.is still in the early stage of development,and many of processes are less practical.In this paper,the extraction processes of alkaloids was researched by studying on seeds of Sophora alopecuroides L.and the separation and purification of the monomer have been also studied to explore a more simple and reasonable technology.So that,the data is acquired to provide a reliable basis for the industrialization amplification.The extraction technology of alkaloids from Sophora alopecuroides L.was studied by microwave-assisted extraction technology,and orthogonal experiment was carried out.The results showed that the microwave-assisted extraction technology was better also,and the optimal conditions were: microwave extracting time was 30s,microwave power was 360W,ratio of solid to liquid was 1:5 (g/mL).The extraction rate of alkaloids was increased by 50%than traditional extraction technology.A reasonable cation-exchange resin used in extraction of alkaloids was screened from varieties of resins.The results showed that the optimal conditions were:concentration was 7.0mg/mL,pH was 3,operation rate was 4mL/min;The effection of elution would be better on condition that elution rate was 2.5mL/min,the pH of ammonia containing 20%ethanol was 11.The alkaloids purified was studied by using the efficient method.After several pre-processing,Matrine and Sophoridine could be separated and acquired by dealing with different pH gradient and solvent.The purity of Matrine and Sophoridine analyzed by HPLC was 98%respectively.
