The contents of metal elements from Capparis spinosa fruits were determined by ICP -AES technique and the flame atomic absorption spectrophotometry (FAAS). The results show that the concentrations of Mg, Ca, Fe, Zn, K, Cu, Sr and Mn are higer than those of other elements.The essential oil of Capparis spinosa fruits was extracted by the method of solvent refluxing and 14 compounds were identified by GC-MS.The content of oligosaccharide was determined by phenol-sulfuric acid method. The effects of time, temperature, solid-liquid ratio, the ethanol solution concentration on the content of oligosaccharide were investigated in orthogonal experiments. The results showed that the sequence of four influence factors were extraction time > extraction temperature > the ethanol solution concentration > solid-liquid ratio in turn. The optimal technological parameters were established: 70℃of extracting temperature, 70min of heating time, solid-liquid ratio1/25, the ethanol solution concentration was 80% .Under this condition, the content of oligosaccharide was measured about 3.70%.The optimal condition for extraction polysaccharide was determined by orthogonal test. The effects of time, temperature, solid-liquid ratio, extraction times on the content of polysa- ccharide were investigated. The optimal technological parameters were established: 60 min of heating time, 80℃of extracting temperature, solid-liquid ratio 1/30, extraction times 3. Under this condition, the content of polysaccharide was measured about 28.72%. The crude polysac- charide was deproteined with Sevag method and the relationship of deprotein times and depr- oteinization ratio was studied.Four ultrafitration membranes in different molecular weight cut-off (MWCO) were used in the isolation of Capparis spinosa polysaccharide. The optimal conditions were as follows: the operation pressure was 0.14 MPa , the concentration of polysaccharide was 0.4 mg/mL. The content of Capparis spinosa polysaccharides in different MW was determined by phenol -sulfuric acid method. Results showed that: 75.52% above 60kDa, 12.32% between 30-60 kDa, 2.9% between 3-6kDa.The coefficient of restitution for pure water flux was used to evaluate the effect of the cleaning. The suitable membrane cleaning agents were: H2O, 0.1 mol/L HC1, 0.1 mol/L NaOH,NaClO+H2O(1/30). The membrane flux could recoveried to 80% of the initial membrane flux.Above 60kDa Capparis spinosa polysaccharide can scavenge hydroxyl free radical of Fenton reaction, and the effect is equal to ascorbic acid and is stronger than that sulfourea.
