The purpose of this study is to solve the problem that organic solvent was used in large quantities and harmful solvent residue in the preparation of silymarin. In this dissertation, influencing factors using alkaline water to extract and isolate silymarin were investigated.This study summarized the optimizing technological conditions and flow sheet, provided experimental reference for industrialization production. The major findings are as follows:The method which to extract and isolate silymarin by ultrasound-alkaline was established. It was shorter time cost, lower energy consumpt, and higher extract efficiency. With the single-factor and orthogonal experiments, the optimizing technological conditions were determined: ultrasound time was 25min, experiment temperature was 30℃, the concentration of NaOH was 2.0mol/L, soak 30min before ultrasound.With dissolved in ethanol and alkaline solution of silymarin, the optimizing adsorbent was S-8 macroporous resin, and the optimizing conditions of adsorption: silymarin saturated solution in alkaline solution (pH13.0), room temperature. The optimizing conditions of eluant: acid solution (pH2.0), 70% ethanol. Influencing factors of S-8 resin dynamic adsorption and desorption wre studied: adsorption rate was 3BV/h, acid eluting rate was 3BV/h and ethanol eluting rate was 3BV/h. At the optimizing conditions, the influencing factors of adsorption about height and diameter of column was studied. Extract-adsorpt composite operation process was determined. |
