| Ergosterol is an important pharmaceutical intermediate.It was producted by two methods,jeast ferment and abstract from antibiotics'mycelium. The key problem existing in the jeast ferment is its high production cost. The environmential problem was solved by abstract method from antibiotics mycelium as well as the high ergosterol production cost is reduced.In this paper, A systematic study on the abstract and crystalization process of ergosterol was performed, and specified saponification, extraction and crystal purify flow.The method of Specification was saponified the wet penicillin mycelium directly. Alcohol and sodium hydroxide were used as saponification menstruum. The optimization parameter: rate of raw material: soponification menstruum was 1:2, rate of alcohol:alkali was 3:1, alkali concentration was 15%, saponification temperature was 85℃. The yield of saponification could reach to 0.53%. The new saponification process is strongpoint: front and back working procedure linking up rational,cost is low,energy consumption is small, easy to operate, contaminating rationally,more proper industrialization produces.Crystallization condition is important for the pass getting the high-purity ergosterol, the crystallization process was improved in this paper by new method. The crystalization condition was optimized by controlled the crystalization concentration to 0.3%, and the cooling velocity to 10℃/h,then got the primary ergosterol.Used ethyl ester and water as menstruum for second crystallization, that would increase the final product content to 99%. After optimization, the yield for ergosterol extraction process was 0.41%.The optimization for ergosterol extraction process would reduce the production cost,raise the extraction yield and quality of final product. At the same time,the research would realize penicillin's cleaner production,solve the environmential problem. |
