Research On The Antioxidant Activity Of Different Solvents Extraction And Purification Of Ergosterol In Hehenbuehelia Serotinal

Hohenbuehelia serotina is one of the famous wild edible fungi in Northeast china.Hohenbuehelia serotina contains a variety of bioactive components such as phenols,flavonoids,polysaccharides,can regulate the immune and anti-oxidation,anti-inflammatory and anti-tumor effect.In order to further explore the Hohenbuehelia serotina in medicinal composition and antioxidant activity,this paper carries out the following research:1.Trace elements and minerals content in Hohenbuehelia serotina were determined by ICP-MS.The results showed that K,Mg,Ca and Fe contents were higher than 100 mg/kg with Hohenbuehelia serotina,Na and Al were higher than 50 mg/kg,which met the GB5009.182-2017 standard;Hohenbuehelia serotina also contains tace elements of a small amount of V,Cr,Mn,Co,Ni,Cu,Zn,Se,Ba etc.;a small amount of harmful heavy metal elements are in line with GB2762-2017 standards of contaminants.2.Using eight different polar solvents of Hohenbuehelia serotina were extracted,and determine the content of total phenolics,flavonoids extract,ergosterol and adenosine yield;and the antioxidative activities.The results show that the different polar solvent in water extract was the highest;eight different polar solvent extraction of total phenol content was higher in liquid water,acetone and anhydrous methanol solvent;the higher the content of total flavonoids of acetone and n-hexane;ergosterol content was the highest butanol;the highest content of adenosine in methanol solvent.Hohenbuehelia serotina extract on the DPPH radical scavenging rate and relative reduction ability is the strongest in the water.3.The extraction of ergosterol by ultrasonic assisted was studied and optimized by response surface method.The best extraction conditions were obtained through the Box-Behnken experimental factor level design:the ratio of material to liquid 1:17,ultrasonic extraction time 55 min,ultrasonic extraction power of 350 W.4.The ergosterol purification with macroporous resin was studied.From five kinds of macroporous resins X-5,D101,NKA-9,D4020,AB-8 were screened for D101 if identified as ergosterol purification;sample concentration of 2.5 mg/mL,sample volume 3 BV,desorption flow rate of 3 mL/min purification conditions of D101 macroporous resin;Determination of extraction with ethyl acetate,the purity was 56.04%to ergosterol;using silica gel column chromatography,obtained 80.30%purity of ergosterol.
