| Penicillin fermentation mycelium waste slag contains abundant organic matter, the organic matter have high extraction using value. Waste slag and production waste water containing residual penicillin fermentation, if no treatment measures were taken to them, they will cause harm to ecological environment.Based on the broken wall, saponification, extraction, crystallization process, ergosterol of penicillin mycelium waste residue can be separated and purified.The wall heat reaction with alkali method, the optimum wall-breaking conditions determined by orthogonal experiments. Wall-breaking fluid solid-liquid separation after reaction, take solid sediment to alcohol alkali saponification process, the optimum saponification conditions determined by orthogonal experiments. After saponification reaction, the liquid extraction by organic solvent, high purity of ergosterol is obtained by distillation and crystallization. Laboratory of extracting ergosterol quality accounted for 0.54% of the dry weight of the residue (the yield of ergosterol was 71.05%), ergosterol purity is 94.2%, conform to the the requirements of as a source of vitamin D2.In this research, the light degradation method and fenton reaction method were used to remove the penicillin of waste residue, by single factor experiment, the most appropriate amount of degradation of material and the reaction of pH were determined, and penicillin degradation effect of three methods are compared, it is concluded that when the reaction time and reaction temperature is fixed, add solvent degradation, the solution pH is 10, the penicillin of waste residue from the initial concentration of 6110 mg·L-1 dropped to 40 mg·L-1, penicillin removal rate can reach 99.35%, and through the degradation products of microbial activity influence experiment found that degradation products had no effects on microbial activity.In this paper the settlement method,method of solid-liquid separation, the method of organic solvent recovery and the method of crystallization of the enlarge experiment for ergosterin extracted amplification experiments is improved,the equipment of magnified experiment made second choice, identified in the process of the settlement of wall breaking liquid add solvent flocculation to improve flocculant sedimentation velocity of solid material, solid-liquid separation of the liquid using the simulation of plate and frame filter method, organic solvent recovery using atmospheric distillation, using organic solvents in the process of crystallization, and repeated use. The extracting ergosterol quality accounted for 0.34% of the dry weight of the residue is obtained by 8 times magnified experiment, the result declined in comparison with the extraction rate of laboratory, achieve the same purity of ergosterol in laboratory. |
