Using of the knowledge of analyze chemistry,Chinese traditiontal medicine, botany etc, we establishes the method that can extract Mangiferin from Rhizoma Anemarrhenae. That method was excellent to turn every kind of parameter, getting a higher putity Mangiferin, wasing authenticated by UV, ER and HNMRs to verify it is a single pure Mangiferin . And was examined by HPLC pure degree over 99%, can be used as standard. Enlarge the technics, we can try out to turn the production in the small scale product.The method provides a way of thinking for extracting anthoer materital .This paper estabilish a HPLC assay for determination of mangiferin . Through the research of methodology,we make sure of chromatogram condition .A kromasil C18 column(250mm*4.6mm 5um) with the mobile phase of acetonitrile-0.05mol/L KH 2PO4(PH3.0),the proportion is 12:88.The velocity of flow is 1ml/min. Examination wave-length is 258 nms.The mangiferin showed good linearity over the ranges of 0.024ug~1.32ug (r=0.9996).The within-day precision RSD was 0.68%.The recovery was 98.9%.The method was easy,rapid,accurate,and sensitive.It will be used in determining mangiferin in herbs .With the method of HPLC, the paper mensurated the mangiferin in Rhizoma Anemarrhenae from different place.The results showd obvious difference for different place Rhizoma Anemarrhenae.This difference was because of its growth environment. We establish th base for evaluating the different place herbs by the fingerprint diagram.The paper determined contents of mangiferin in Rhizoma Anemarrhenae before and after processing.The contents of mangiferin in Rhizoma Anemarrhenae was rised.This may be the similar to mangiferin transformation to mangiferin . Certainly,must make further research to confirm.Finally, the paper determined contents of mangiferin in Baihu soup. The contents of mangiferin in Baihu soup with crude gypsum was rised,and contents of mangiferin in Baihu soup with processed gypsum has not evidence change .When the proportion of Rhizoma Anemarrhenae and crude gypsum is different,the contents of mangiferin in Baihu soup is different.The contents of mangiferin is the most highest ,when the proportion is 8: 3. This proportion nears to ancient proportion . from this aspect we can see that the ancient proportion has some rationality.The paper establishes a method that can extract Mangiferin from Rhizoma Anemarrhenae and estabilishs a HPLC assay for determination of mangiferin. And use this HPLC method determined contents of mangiferin in Rhizoma Anemarrhenae ,include that different growth plcace herbs and processed herbs,and applied to research the mangiferin in the Baihu soup.
