Technics Of Separating Hongmaoxinjian From Radix Caulophylli. And Its Pharmacy Study

Radix Caulophylli, used as local herb medicine, is the dry root and rhizome of Leontice robustum (Maxim.) Diel (Berberidaceae), only occuring in Qinba mountain areas in Shaanxi. The extract of Radix Caulophylli has many pharmacological effects such as decreasing blood pressure, exciting uterine smooth muscle, antibiotic, antiphlogosis, antivirus, antirheumatism and antispasmus etc. The study on the active components in Radix Caulophyll by cell membrane chromatography with pharmacological experiment resulted in the identification of hongmaoxinjian, and proved that hongmaoxinjian has preventative and therapeutic effects on cardiovascular disease.In order to prepare hongmaoxinjian on large scale, we studied the extracting and isolating technics of hongmaoxinjian from Radix Caulophylli total alkaloids. Firstly, the isolationg effectiveness of hongmaoxinjian on six resins: Macroporous resin XDA-2, AT-32, AT-30 and ion exchange resin B108, AT-50C, LSD001, were examined. The results showed that AT-32 had the best isolating effectiveness. And then, based on the use of AT-32, the isolating and purifying method of hongmaoxinjian was established, which is twice AT-32 absorption treatments, and then silica gel column chromatography, and finally recrystallization. Meanwhile, HPLC analysis method on hongmaoxinjian in Radix Caulophylli total alkaloids was established.Physicochemical properties and stability of hongmaoxinjian were studied to develop a new medicine in future on the base of hongmaoxinjian as acitive component. The results showed that light, heat and humidity made a bit slight decrease effects on the content of hongmaoxinjian. The water-solubility of hongmaoxinjian salt chloride is better than its sulfate and phosphate.For scientifically evaluating the quality of Radix Caulophylli as herb medicine, qualitative analysis methods on hongmaoxinjian and taspine by TLC and quantitative analysis methods by CZE were established, resp.. And sequentially the contents of hongmaoxinjian and taspine in Radix Caulophylli were determinated, 3.8 mg/g and 4.3 mg/g, resp., by means of the established method above.The results in this study provided an important foundation for further deeper research of hongmaoxinjian and its pharmaceutical development, and a dependable analysis methods on hongmaoxinjian and taspine in Radix Caulophyll.
