Objective Study on the extraction, purification and quality standards of daphnoretin and total flavones,which are the major active components of Radix Wikstroemiae Indicae,so as to obtain high purity products.Furthermore,Study the quality standards of the daphnoretin and total flavones in order to get a good quality control.Methods Using the content of daphnoretin and total flavones and extracting ratio as evaluating indexs,we selected the optimum extracting condition by orthogonal; purify the extract of Radix Wikstroemiae Indicae in order to get a higher purity product, and the daphnoretin in the extract were investigated by HPLC and total flavones with visible spectrophotometry.Done by TLC Identification;using High Performance Liquid Chromatography (HPLC) determination, visible spectrophotometry and methods of inspection. Results The optimum extracting condition was:55% ethanol immersion 0.5 hours,heating to extract two times,adding alcohol for 10,8 times and extracting for 1.5,1.0 hours. The technological conditions of adsorption and desorption for Radix Wikstroemiae Indicae were as follows:the concentration of extract 1.Og/ml polyphenol sample volume is 1.0 times of the bed volume(BV), and a flow rate of 2 BV/h at room temperature; Diameter and height ratio is 1:3.Then use 3 times BV of distilled water for purification whereas the optimal desorption conditions are found to be a flow rate of 8 BV/h using 5 times BV of 80% ethanol for elution. Radix Wikstroemiae Indicae were indentified by TLC. Daphnoretin and total flavones determination feasible,good repeatability.Conclusion The procedure was reasonable and pratical,the quality standards was set up.
