In beings progress,sugar as a energy substance(such as glucose, starch) and structure substance (such as fibre, chitosan et al) , have been known by human being Recently,fungal polysaccharides have been discovered to exhibit many bioactivities, such as anti-tumour, antiviral, anti-Inflammation, hypoglycemic, hypotensive and so on.There are good immunomodulators to inhibit the tumor cells growth with little damage to normal cells.The chemical modification can enhance bioactivities of natural polysaccharides, and even give new bioactivities. It has been reported that sulfation of polysaccharides affect strongly on their immunocyte activity and anti-tumour bioactivities, but the sulfatation of polyporus umbellatus polysaccharides has not been reported.The thesis synthesize the sulfate polyporus umbellatus polysaccharides for the first time. It is a great innovation.The main contents in this thesis are divided into the following parts.1. By the method of Orthogonal experiment, optimize the technics of extract. Water-soluble Polyporus umbellatus polysaccharides was isolated successfully from the fruiting bodies of Polyporus umbellatus. Weight-average molecular weight (M_w) was determined by using size excluded chromatography (SEC) combined with static light scattering (SLS) to be 1.6×10~5Del.2. Use the method of phenol-vitriol determine the content of general-sugar is 83.4%, By the method of Gas Chromatography (GC) analytic, analyse the monosaccharides of pps-1, the result showed that pps-1 was mainly composed by D-glucose with a few D-galactose and D-mannose.3. Sulfated with the method of chlorosulfonic acid-pyridine. Through add in dispersant DMF to analyse the result of sulfation.The structures of sulfated polysaccharides was determinated by FI RD analyse, After adding dispersant, the yield of production of sulfated polysaccharide increased, but it reduced the molecular weigh and the content of sulfate group.4. The results showed that pps, ppss-1, ppss-1' solutions at extremely low concentration displayed Newtonian flow behavior at 25℃.5. After added dispersant, the production of sulfated polysaccharide shows exhibit well bioactivities by immunity experimentation, the immunocompetence has been great enhanced.The results of foundation research mentioned above provided a lot of scientific...
