Soluble soybean polysaccharides (SSPS) is the water-soluble polysaccharides extract from the cotyledons of soybean. SSPS is one of the effective ingredients in soybean, it could have been made into high concentration solution, and the solution viscosity influenced little with the salts. It also more stable than other carbohydrates. SSPS have various functions such as antioxidant activity, stabling protein particles in acidic solutions, decentralized nature, emulsion, adhesive, etc. It can not only be used as Dietary Fiber, it also can be used in industrial.In this paper, we researched on the extraction of soybean polysaccharide (ESP), then purification and analyze its monosaccharide components. The ESP primarily focuses on the quantities of SSPS under different temperatures, time, pH, as well as conditions of varied firm than liquid, thus finds out the best arrangements of the four factors, namely temperature 100℃, time 3 hours, pH 5, and firm than 2 liquid. The SSPS extraction under the best conditions would be separated by DEAE-52 cellulose column chromatography. We got the largest eluting peaks by using double-distilled eluted. The concentration is about 70.58%. After the elementary purification, the extraction is identified with TLC (Thin Layer Chromatography) and FTIR (Fourier Transform Infrared Spectroscopy) for its monosaccharide constitutions. The result is that in our experimental conditions the monosaccharide components of SSPS is rhamnose and galactose, and so on.Research shows that polysaccharide have the immunologic-regulation effect. Massive immunity tests proved that the immune regulation of polysaccharides is one of the most important roles. In this paper, we use animal experiments, to observe its immune activity. Index of immune organs was weighed and calculated. Phagocytes of mononuclear macrophage were determined with carbon particle clearance test. Peritoneal macrophage phagocytes also be tested. The results showed that high doses of SSPS can effectively improve the mice immune function. The carbon particle clearance experiment got the middle dose group maximum. This may be caused by the difference of mice weight. The result of the tests is that SSPS have the immunologic-regulation effect.
