Galanthamine contained in the bulb of amaryllidaceae is a specific, emulativeand reversible acetylcholinesterase inhibitor, which is effective on the treatment ofmild or moderate ischemia agnosia of Alzheimer's disease patients.In this paper, supercritical carbon dioxide extraction was used to extractgalanthamine from lycoris radiate.Firstly,microscopic identification with starch grain was used to identify theextent of cell wall fragmentation in the bulb of lycoris radiate that was pretreated bycomminution and ultrasonic comminution. The results indicated that the cell wallcould be broken up by comminution and ultrasonic comminution pretreatment. Theeffect of cell wall fragmentation by ultrasonic comminution was more obvious thanthat by comminution. Moreover, the results of SFE showed that the operation timewas shortened and the yield of galanthamine was increased after ultrasoniccomminution pretreatment.Secondly, the split phase point track of CO2/ethanol/water ternary system wasmeasured by the research of phase equilibrium of the system, which providedguidance for the choice of entrainer and process parameters in the extraction ofgalanthamine by supercritical carbon dioxide.According by, 90 wt % of ethanolsolution was determined as the entrainer and the flow rate was 5ml/min.As addition, the effect of each factor on the yield was investigated by orthogonalexperiments. And it was concluded that temperature was the most influencing factor,then pressure, CO2-flow rate in turn. The optimal conditions of supercritical CO2extraction were as follows:temperature, pressure and CO2 flow rate were 65℃,40Mpa, 20g/min respectively, under which conditions the yield reached 1.45%00. Andcompared from the yield and selectivity, SFE is preferable to traditional organicsolvent extraction.The problems such as complicated operating steps, large discharge capacity andunavoidable solvent residue limited the further application of. the traditionalextraction of galanthamine by organic solvent. The new process of SC-CO2 extractiongalanthamine from lycoris radiate with SC-CO2 gave some references to extract otheralkaloids.
