Selective supercritical fluid extraction (SFE) of glycolipid from wheat flour used prior to chromatographic determination allows reduction of sample preparation time from 17 hours to 30 min. Practical quantitation of certain lipids, particularly digalactosyl diglyceride (DGDG) related to breadmaking of wheat, can be useful in the evaluation of new cultivars. Quantitation of the extract by supercritical fluid chromatography (SFC) also made the determination possible in 10 min. compared to the 80 min. previously developed high performance liquid chromatography (HPLC) procedure.
