| The dispersible tablet is a dosage form that release itself quickly. This kind of dosage form have some merits of both tablet and suspension and increase bioavailabilityThe formulation is composed of three traditional Chinese medicine. Danggui and Chuanxiong belong to the same department and can be extracted together. The optimization extract method was determined by orthogonal design; the ratio of solvent (85% ethanol) and material is 8:1, and extract two tunes, each time is one and half an hour. Gansong was heated for four hours to extract volatile oil and add water 10 times to boil it another one hour.The saturation solvent technique was selected in the preparation of β-Cyclodextrin inclusion containing Gansong volatile oil. In results, the optimized process is as follows, the ratio of Gansong volatile oil and β-Cyclodextrin is 1 to 10, inclusion temperature was 80癈 for one hour.The Compound Gansong dispersible tablet was prepared by direct compression. The choice of disintegrants was studied by orthogonal design. The results show that MCC was 40%and CMS-Na was 6%. The prepared tablets were concluded Ferulic acid 0.353mg each tablet. The test of disintegration and dissolution was conducted based on Chinese-5-Pharmacopoeia (2000).Healthy rabbits were used to study preliminary pharmacody -namics of Compound Gansong dispersible tablets. The results of research indicated that coagulation of blood platelet was significantly inhibited by the Compound Gansong dispersible tablets and its extract method. While Cuanxiong and Danggui deposited in 70% ethanol after extraction in 85% ethanol, the inhibit rate (IC50) was 10.92mg/ml and most effective.The pharmacodynamics of the Compound Gansong dispersible tablets was performed on Kunming mice and Wistar mice. The results show that compound Gansong dispersible tablets can prolong the time to form thrombus after electric stimulation carotid and obviously restrain to coagulation platelet cruor and prolong cruor time. |
