The technology of light induced fluorescence and fluorescence imaging are used widely on the diagnosis of cancer because of the characters of on time, mini-invasive or non-invasive, low side-effect and high sensitivity. Cancer is called as well as malignant tumor, refers to the abnormal hyperplasia of local tissue cell induce local lump at the effect of tumorigenic facts. Malignant tumor can destroy the structures and functions of tissue and organs. The patients may die as a result of the functions of organs failure in the end. So it is important to diagnose cancer early.This paper is based to fluorescence spectroscopy diagnosis technology, analysing and contrasting the spectra characters of different photosensitizers. Combined with clinical requirements, Rose Bengal is chosen as photosensitivity drug to diagnose oral precancer. The best excited wavelength is decided by experiment.This paper applys the technology of light induced fluorescence to design and establish the system of fluorescence spectroscopy diagnosis. The system consists of LED with the midwavelengh 405 nm as to be excited lamp-house and USB 2000 monochromator as to be measure apparatus in order to measure on time. The golden hamsters are used as experiment objects, Rose Bengal is used as photosensitizer. A parameter Q is calculated by fluorescence ratio method by analysing the fluorescence spectroscopy characters of precancerous different stages. Oral precancer can be diagnosed on the basis of the parameter Q .The result shows normal oral mucous membrane tissue can not be dried by Rose Bengal, but the lesions and cancers tissue can be dried. After topical application of photosensitizer, characteristic emission fluorescence is observed and it is around 570 nm to 600 nm. Similar, normal oral cavity has special luminescence around 480 nm. When the malignant tumors appear, the characteristic fluorescence spectra change. Typical porphyrin peaks appear at 630 nm and 690 nm. Fluorescence imaging diagnosis can be applied on the basis of the character of photosensitizer. It is helpful to tumor localization.
