Worldwide,esophageal cancer with a high mortality is the eighth common cancer.The principle of treating cancer is that early detection,early treatment.Early esophageal cancer can be cured by surgical resection and has a good prognosis.However,due to the limitations of current clinical diagnostic techniques and people's insufficient understanding of esophageal cancer,early esophageal cancer is difficult to be found.When the patient is diagnosed,the cancer is often in the advanced stage.Different stages of cancer need to take a different treatment options.In the advanced stage of cancer patients,the survival rate dropped significantly.Therefore,in order to improve the survival rate of patients with esophageal cancer develop a new technology used for accurate diagnosis of early esophageal cancer has become the key scientific problems to be solved.This paper,by using multiphoton microscopy(MPM)imaging techniques for early esophageal cancer diagnosis experiment,aims to illustrate the clinical application of this technology in the diagnosis of early esophageal cancer potential.First,the use of MPM to detect multiphoton signals in stained tissues.The results show that both SHG and TPEF signals are present in both stained and unstained tissues.Second,using MPM imaging the normal esophageal tissue,we demonstrated that the MPM has the ability to present microstructures of the four-layer structures of the human esophagus wall.Finally,MPM imaging of H&E-stained esophageal frozen sections achieved during surgery were performed.Our results indicated that MPM has the comparable ability with H&E-stained not only to clearly reveal biological tissue microstructure and its morphological changes and identify the tumor cells,distinguish between normal tissue and tumor tissue but also can reveal information not distinguishable in H&E-stained image under classical bright field approaches.The complementary information of nonlinear spectral analysis of the normal and cancerous tissues,quantitative describe the changes of collagen fibers in the normal and around cancerous tissues showed that MPM has important accessory diagnostic value for the differential diagnosis of submucosal cancer of esophagus during intraoperative period.
