Clematis Tangutica(Maxim.) Korsh.growing in the northeast of Qing-Tibetan Plateau was selected as the experimental material in this thesis.The growth rhythm and botanical characteristics of Clematis Tangutica(Maxim.) Korsh.were researched using field spot-observation method;The community quantitative characteristics of Clematis Tangutica (Maxim.) Korsh.in different regions were analyzed using the least sample method;Xining was the sample plot,the introduction and acclimatization test including seedling,cottage reproducing, transplanting showed positive results.Thin Layer Chromatography(TLC) was used to identify oleanolic acid;both indices of HPLC content determination and HPLC fingerprint spectrum were used to conduct the quality control standard.The result showed:(1) Clematis Tangutica(Maxim.) Korsh.distributed at an altitude of 2370-4000 M of narrow areas in Qinghai Province,such as,Haixi,Haibei,Hainan,Huangnan, Yushu,Guoluo districts and the east agricultural areas.(2) In the main distributed area,both dominant species and constructive species of plant community were Clematis Tangutica(Maxim.) Korsh.and the companion species was mainly Poa Annua L.(3) The plant phenological period of Clematis Tangutica(Maxim.) Korsh.in Xining was divided into six stages,,namely,Germination and Emergence stage,Young seedling stage, Growing stage,Flowering stage,Filling and Withered stage.(4) The over-winter storage,water soaking treatment in variable temperature,different concentrations of KNO3 and GA3,variable temperature treatments,all could improve the germination rate;the most optimal treatment composition was over-winter storage with -10℃and treatedwith ppm GA3,seedling at constant temperature,the germination rate would reach 73%.(5) The germination rate of Clematis Tangutica(Maxim.) Korsh.was 3%and the survivability of transplanting was 50%and the survivability improved dramatically after cutting brunches was treated in indoleacetic acid(IAA).(6) TLC technique was used to extract chloroform from Clematis Tangutica(Maxim.) Korsh..this technique was a quick,simple and convenient evaluation method for the quality of Clematis;The TCL result showed that Oleanolic acid was existing,the constituents might support the Clematis on chemotaxonomy. (7) HPLC fingerprint of Clematis Tangutica(Maxim.) Korsh.was primarily identified,it indicated 15 common-used chromatographic peaks and 12 non common-used chromatographic peaks;HPLC fingerprint combined with content determination of oleanolic acid were the quality control method of Clematis Tangutica(Maxim.) Korsh.,this method also could be applied to other medicinal plants as reference.The innovation points of this thesis were following:(1)For the first time,systemically studied the medicinal plant- Clematis Tangutica (Maxim.) Korsh.growing in the northeast of Qing-Tibetan Plateau.It provided the practical and theoretical basis on the reasonable utilization,development and protection of Clematis Tangutica (Maxim.) Korsh.,a kind of medicinal plant in Qing-Tibetan Plateau.(2)For the first time,the acetonitrile- phosphate technique was used for the HPLC fingerprint research,and two identification methods were used to conduct the quality standard; accumulated practical and theoretical experience in Clematis quality evaluation and classification.
