An experiment was conducted to analyze the phenotypic diversities in terms of 6 phenotypic traits(height of plant ,length of petal,number of leaf,number of flowers ,length of leaf and width of leaf)using 7 natural populations of Lilium regale Wilson. Analysis of variance for all traits showed that there was significant difference among populations. Among populations, the F value of 6 traits was from 4.9 to 34.9, except leaf was significantly different; other traits were quite significantly different. Within population other traits were not significant except that length and width of leaf. The main phenotypic differentiation coefficient for the 6 traits is 61.5%, and the variation among populations was higher than within population, which indicated that the variance among populations was the main part of the phenotypic variation. The correlation analysis between the phenotypic traits of Lilium regale Wilson and geography factor showed that the correlation between them was not significant except that height of plant and the number of flowers and width of leaf significantly different positive affected by latitude.7 natural populations of Lilium regale Wilson could be divided into 2 groups by UPGMA cluster analysis. Explaining the phenotypic traits didn't gathers a type according to geography distance.Lilium siberia were used to study meiosis and male gametophyte development using staining and chromosome preparing methods.The result showed that: six anthers, four locules. Cytokinesis of Lilium siberia belonged to successive type. The microspores in tetrad stage showed the arrangement of decussate type and tetrahedron. Mature pollen grain was two celled type with one germinal furrow. There were two to four division phases simultaneously observed in the same plant and same flower buds and even in same anthers and this meiosis outasync was of important significance in population reproduction and propagation of the genus.The exceptional meiosis of Lilium siberia pollen mostly happened in every period of microspore mother cell meiosis and microspore monokaryotic forepart, it is important reason that result in male sterility.Lilium siberia pollen storage in 4℃, after five days, the pollen germination evidently descend. It is indicate that pollen storage endurance is weak. Observed Siberia lilium pollen bourgeon and pollen tube growth on the stigma and cut-style with fluorescence microscope. The result indicated pollen tube stop to grow in the style, couldn't arrived in ovary accomplish spermatize, so distant hybridization can't appetency, during of haploid inducement, the method of farina growth is multiform.
