DNA molecular marker is a new adwanced technology applied in molecularbiology and biotechnological research.It has some particular advantages compared with other markering technology,such as morphologic marker,cytological marker and biochemistry marker.At present,there are five major methods, RAPD,SSR,AFLP,RFLP and ISSR used to detect the DNA variance.Now,the technology of DNA molecular markers are being widely used in the areas of plant research.Suaeda salsa,belonging to Suaeda of Chenopodiaceae,is an annual herb.S.salsa has the character of strong resistance,easy propagation ,long life of seed.It can survive in 4.3%sea water.At present,the using work of S.salsa is lack,especially the theoretical guidance on genetic diversity research.This study selected 80 individuals of 8 populations of S.salsa which natual distributed in differet salinity soil in Dongying ,Shandong and adopted SSR and AFLP as the molecular markers to detect the genetic diversity .It indicated that there existed the correlations between gengetic differentiation and geographic distance in S.salsa population. An understanding of both genetic diversity and population structure of S.salsa provides insight for the conservation and management of S.salsa.This study obtained the conclusion there in after:1.The genetic diversity of S.salsa revealed by SSR and AFLP on species level:(1)45 pairs of primers had been used to analyse the genetic diversity of 8 populations of S.salsa.Each of 16 pairs of primers generated 2-6 loci with 4 loci in the average.All of the primers totally 65 loci were generated and 48 loci had polymorphism which occupied 73.2%. The polymorphism information content among the 16 pairs of primers were analyzed,primer 6 got the highest PIC 0.5143, primer 13 got the lowest 0.3385. The D of 8 populations located between 0.0747-0.1997.(2).40 pairs of primers had been used to analyse the genetic diversity of 8 populations of S.salsa. Each of 6 pairs of primers generated 23-49 loci with 37 loci in the average. All of the primers totally 223 loci were generated and the loci that had polymorphism occupied 70.3%. The polymorphism information content among the 6 pairs of primers were analyzed,primer 2 got the highest PIC 0.3596, primer 4 got the lowest 0.2014. The D of 8 populations located between 0.0497-0.2071.All of these two molecular markers are the suitable one to test the genetic diversity of S.salsa.2.Comparison between SSR and AFLP: Loci polymorphism ratios of SSR and AFLP were73.2%and 70.3%respectively.Each primer could generate 4 and 37 loci for SSR and AFLP.According to the cluster analysis derived from our experimental results by the two techniques,both can analyse the genetic diversity of 8 populations of S.salsa,but they have some differences,and there are some relations between the genetic diversity and concentration of salt.
