To know halophilic actinomycetes and their enzyme-producing functional diversity from mud volcano by cultured approach in Usu county, Xinjiang. Soil samples in mud volcano were isolated using five isolation media containing 5% and 10% NaCl with dilution-plate method. The enzyme activity was qualitatively detected by using five selective media. Base on morphological characteristics, test of salt tolerance, screening of enzymology characters and sequencing of 16S rRNA gene , strains were selected for phylogenetic analysis. A total of 43 halophilic actinomycetes and 3 extreme halophilic actinomycetes were obtained in our study. Screening results for enzyme activity showed that 4 halophilic actinomycetes produced lipase, 30 strains produced galactosidase, 27 strains produced amylase, 6 strains produced esterase, 4 strains produced cellulose and one strain produced four enzymes simultaneously. Of these, 30 strains were selected for homology and phylogenetic analysis of 16S rRNA, which confirmed that 29 strains were belong to Nocardiopsis and one strain was Streptomyces. Strain 10006 producing two enzymes and Nocardiopsis exhalans (AY03600)showed 96.64% similarity (less than 97%), which indicated that it was a new possible species. Totally 65 strains of bacteria were isolated from mud volcano samples,it were highly related to the known bacteria in the genus Staphylococcus, Alkalibacterium, Planococcus, Brachybacterium, Marinobacter, Halomonas, Salinicoccus, Nesterenkonia, Bacillus.To know bacterial diversity from mud volcano by uncultured approach in Usu county, Xinjiang. The mixed environmental genomic DNA was directly isolated from the mud volcano in Usu county, Xinjiang. The 16S rRNA genes were amplified with bacterial universal primers and cloned into the pGEM-T Easy Vector, resulting in the construction of a16S rRNA clone library.The total of 150 clones were selected by amplified rDNA restriction analysisusing restriction enzymes Haeâ…¢. Of these, 16 restriction fragment length polymorphism(RFLP)types were detected and sequenced. Most of the cloned sequences were highly related to the known bacteria in the phylum Proteobacteria,Firmicutes,Fusobacteria,Actinobacteria (>96% sequence similarity),while small part of the cloned sequences showed less affiliation with known taxa (<94% sequence similarity) and may represent novel taxa.
