High hydrostatic pressure (HHP) regarding proteins, nucleic acids, lipids and polysaccharides influence mainly their structure change, the pressure on these biological macromolecules in effect and mechanism is different from the temperature and other chemical factors. So far, the impact of high pressure to cell is little researched. Actually the pressure has important influence to the cell metabolism and gene expression. Pressure on the molecular interactions can help us to get a better understanding of the pressure on the cell biological systems. The new research data indicated we have not been able to expect the outcomes of the interaction between the protein and the nucleic acid, however it also shows we should grasp some new theories to better understand the metabolic system. of cell biology.MicroRNAs(miRNAs)are a class of highly conserved endogenetic noncoding RNA with length of ~22nt. The miRNAs silence its target genes in post-transcriptional level and play roles in almost all physiological process for example development, differentiation, proliferation, apoptosis and pathological changes. The miRNAs express at specific space and time strictly. The abnormal expression of miRNAs will result many diseases such as development defects, cancer, obesity. The miRNA expression profiles are very helpful for us to understand miRNA functions. So far, the microarray is the most powerful method to study miRNA expression profile high-throughly.This experiment first carries on high hydrostatic pressure processing to mouse teratoma cell P19, through exploration of pressure conditions, finally in 120Mpa, 20min, 37℃we obtain the pressure resistant P19 cells, in order to further researching its molecular mechanism, using RT-PCR technology and chips to build miRNAs expression profiles, then study miRNAs expression of the pressure resistant cells. Using this method we have analysed miRNAs expression differences between normal raise P19 cells and pressure resistant P19 cells and discovered several obvious expression differences.
