Chiral the existence of the natural world is a universal phenomenon. Organisms constitute the basic material, such as amino acids, proteins, nucleic acids, carbohydrates and other biological molecules are chiral molecules. Apart from a few exceptions, almost all protein is the L-amino acids, polysaccharides and nucleic acid in sugar is D structure. Related to this many drugs, physical substances, pesticides and herbicides are also optically active compounds. When it go into the body, its pharmacological and physiological role relate with the match of target molecule between molecular recognition and ability. Chiral 2-octanol is one of the most important materials of liquid crystal and liquid crystal devices, and its optical rotation and high dielectric constant and most of LCD performance are closely linked.Enzyme as a biological catalyst can be used for the synthesis of optically active compounds and resolution. Because of its high enantioselectivity, little vice reactions, therefore we can get high yield and optical purity. In addition enzyme-catalyzed reaction can be reacted under mild conditions and pollution-free.While non-aqueous phase catalytic reaction is convenient, but the traditional organic solvents are usually restricted enzyme activity and selectivity. These the years, people have been trying to solve the problem. In recent years, a new attractive non-aqueous reaction medium for biocatalysts has emerged. Ionic liquids, because of their negligible vapor pressures, have been generally recognized as green solvents. Use of ionic liquids as media for performing biocatalyst reactions has gained increasing attention, with sometimes-remarkable results. Ionic liquids are organic salts, which are liquids at ambient temperatures. Unlike traditional solvents, which can be described as molecular liquids, ionic liquids are composed of ions. Their unique properties such as non-volatility, non-flammability, and excellent chemical and thermal stability have made them an environmentally attractive alternative to conventional organic solvents. Ionic liquids have low melting points (<100℃) and remain as liquids within a broad temperature window (<300℃). Because of the high polarity, ionic liquids present an ideal reaction media for chemical and biochemical reactions due to their ability to dissolve a wide range of different substances including polar and nonpolar organic, inorganic, and polymeric compounds.Magnetic microspheres are a kind of microspheres which can be made by an appropriate way to join organic polymer with metals or metal oxides ultrafine powder of magnetic properties. This magnetic microsphere has two notable features: First, it's super-paramagnetic. In the magnetic field show the presence of magnetic, when the magnetic field when the withdrawal is rapidly disappearing; Second, the characteristics of a polymer particles can surface chemically modified to give the surface a variety of functional groups. It is precisely because of these two aspects of the characteristics, magnetic microspheres show a strong vitality in many areas.In this paper, we prepared tetrahydrothiophene ionic liquid, by changing the alkyl chain length to adjust the cation, and select I-, N(CN)2-, N(SO2CF3)2-and PF6- as anion, in order to prepare low-cost, higher yield, high purity, low viscosity room temperature ionic liquids. Their applications in the PSL lipase catalysis resolution 2-octanol were also researched. We also found different cationic, anionic of ionic liquids as a catalyst additive on the impact of resolution. Hexafluorophosphate S-ethyl tetrahydrothiophene was found the best ionic liquid addictive in this resolution. The best concentrate of ionic liquid in this resolution was also found. Compared with BMIM·PF6, which is the normal ionic liquid used in biocatalysts, S-ethyl tetrahydrothiophene perform a higher reaction activity and E value.Since there were some problems about the recover of enzyme in ionic liquid, we prepare so we prepared 15 nm, monodisperse magnetic microspheres, which containing free amino or carboxyl. It can be coupled with the PSL containing free amino and carboxyl .The immobilized enzyme is paramagnetic, and can be separated and removed after the reaction by a magnetic.We also examines two kind of immobilized PSL in ionic liquids hexafluorophosphate S-ethyl-tetrahydrothiophene and BMI·PF6 for the resolution of 2-octanol. Their reaction activity and E values were compared to select a suitable immobilized PSL for this resolution. The reuse cycle of immobilized enzyme were also examined. The reaction activity and E value don't have many changes.
