Seven species of Stipa L. were studied, located in meadow steppe, pink steppe, desert steppe and desert.The sequences about 1120 bp of aquaporins gene of seven species were firstly cloned and sequenced(GenBank Accession: EF061925-EF061931).The structure of sequence, the composition of nucleotide and the molecular evolution trees were analyzed by biology software. The polymorphism of exon1 and intron1 of aquaporins gene were Invested by the PCR-SSCP analysis. The results were that, AQP gene of Stipa L. had three exons and two introns, and the size and the location were high conservatism. Exon1 had three genetype (AA,BB,CC), intron1 had four genetype (DD,EE,FF,GG), and polymorphism existed in species,while no polymorphism were found within species. The composition analysis of amino acid showed that the contents of polarity amino acid was higher than that of apolarity, the contents of positive amino acid was higher than that of negative. From the molecular evolution trees constructed by different tree-building methods, three trees were similar, the evolution species of Stipa L. AQP gene were conformity with geographic distribution of Stipa L. species.
