Microenvironment balance in respiratory system,especially water balance is vital to maintain its normal physiological functions.There are AQP1,AQP3,AQP4,AQP5,AQP8 and AQP9 six members of aquaporins expressed in respiratory system,and AQP1,AQP3, AQP4 and AQP5 are distributed throughout the nasopharynx,airways,and distal lung. AQP1 is mainly expressed in microvascular endothelia and pleura.AQP3 is mainly expressed in the basolateral membranes of basal epithelial cells of large airways.AQP4 is expressed in apical epithelial cells of airways.AQP5 is expressed in the apical membrane of typeâ… alveolar epithelial cells and at the apical membrane of acinar epithelial cells in submucosal glands.The larynx is not only the portion of the respiratory tract,but also contains the vocal cords which produce vocal sound.The regulations of liquid secretion and water balance of larynx are very important for its normal physiological function.There are many reports about the potential roles of aquaporins in respiratory system and some relative diseases at home and abroad so far,but little literature is known about the location and function of aquaporins in larynx.There is only one report showing the expression of AQP1 and AQP4 in rat laryngeal tissue at home.In the present report,we are aimed to study the expression and location of aquaporins in human and mouse laryngeal tissue,then to explore the potential roles and mechanisms of aquaporins in normal laryngeal tissue and relative diseases.We mainly investigated the expression and location of aquaporins in human and mouse laryngeal tissue by using RT-PCR,immunohistochemistry and Western Blot.We found that:AQP1 and AQP3 were expressed in human larynx,and AQP1 was located on the cilia of pseudostratified ciliated columnar epithelium of laryngeal ventricle and on the capillary endothelial cell of laryngeal mucous lamina propria;but AQP3 was expressed on the stratified squmous epithelium and laryngeal lamina propria mucous glands.In mouse larynx,there were AQP1,AQP3,AQP4 and AQP5 expressed,and AQP1 was expressed on laryngeal cartilages cells,AQP3 and AQP4 were expressed on the stratified squmous epithelium of laryngeal mucosa,AQP5 was expressed on the pseudostratified ciliated columnar epithelium of laryngeal mucosa and on the submucosal glands.This study has only investigated the expression and location of aquaporins in human and mouse laryngeal tissue at present.Further detailed studies are needed to reveal the functions of aquaporins under the condition of the laryngeal physiology and pathology.It is very important to study the regulation mechanism of every member of aquaporins and their potential roles in normal laryngeal tissue and relative laryngeal carcinoma,all these studies will provide new useful information for the pathogenesis mechanism and diagnosis and treatment of laryngeal disease.
