Objective: To modify preparation method and to study immunological competence of sheep placenta peptide (SPP). Methods: Sheep placenta peptides were prepared by broking-repeated freezing and thawing method and the optimum preparation conditions were studied. Sheep placenta peptides were intraperitoneally injected to Kunming mice, and indices of immune function were then measured. The cellular and humoral immune function of mice was determined by T and B lymphocytes proliferation and carbon granule clearance test, immuneenhancement effect of sheep placenta peptides was tested using immunologic indexes such as phagocytic index and phagocytic coefficient. T and B lymphocytes proliferation was tested with MTT assay, and the immunological competence of sheep placenta peptides was examinated by MTT assay and carbon particle clearance test. Result: Optimum preparation conditions are as follows: freezing temperature: -30℃: freezing time: 48h; repeated times: 4. Sheep placenta pcptides could increase T and B lymphocytes proliferation induced by ConA and LPS (P<0.01) and enhance immunity function of T and B lymphocytes (P<0.01). The clearance index was obviously reduced by cyclophosphamidun, but sheep placental polypeptides could recover the macrophage phagocytic index and coefficient of mice with immunologic hypofunction (P<0.01).Conclusion: SPP can enhance cellular and humoral immune function and phagocytic function of reticuloendothelial system in mice. Sheep placenta peptide is a reasonable immunomodulator with immunoregulatory activity.
