In recent years, the effect of environmental factors on the growth of biology is one of the hotspots in biological area. Magnetic field, as a kind of physical factor of environment, has great effect on organism, which has become an important content in biophysics. The researcher in different countries has not made consistent conclusion in independent research because of the existence of many uncertain factors such as magnetical strength,action period and material.In this thesis, Polygonatum cyrtonema Hua was used as experimental material, the medium(hormone concentration,carbon source) was optimized to accelarate the growth of rhizome of Polygonatum cyrtonema Hua, then insure the abound supply of materials;After the confirmation of the best treatment method. the Polygonatum cyrtonema Hua plantlets were treated with different magnetic intensity and time. Then the growth condition of Polygonatum cyrtonema Hua was observed, the exterior shape,physiological and biochemical character and the activity of protective enzymes of Polygonatum cyrtonema Hua plantlets were also been discussed.Then the mutation mechanism of plants treated with magnetic field was acquired. the results are as follows:1. The Polygonatum cyrtonema Hua plantlets was cultured under four kinds of NAA concentration and three carbon sources with different concentration. The results showed that the rhizoma required lower concentration of NAA than bud, The multiplication of root was in favor of lower concentration of NAA, and the multiplication of bud was in favor of low concentration of NAA; the concentration of carbon sources could also influence the growth of cultured seedlings, high concentration is better, but there was no diference among the cultured seedlings about the purity of sugar, so sucrose could be replaced by white sugar as carbon source.2. The cultured Polygonatum cyrtonema Hua plantlets was developed in magnetic fieldby three ways. The result shows that the best method is only to treat the cultured seedling.3. After treated with magnetic field, the fresh weight of rhizome of cultured Polygonatum cyrtonema Hua plantlets have increased obviously compared with the control group. The trend of its development was increasing slowly then decreasing sharply. The treatment with magnetic field was propitious to the growth of fibrous roots .after treating the number,length and diameter of fibrous roots all inseased. 4. The treatment of magnetic field on the cultured Polygonatum cyrtonema Hua plantlets could increase the content of chlorophyll,soluble protein and soluble sugar. And those parameters increased to the highest level with all 15 hours, 12mT,17mT,27mT in each group.5. The effect of magnetic treatment on the cultured Polygonatum cyrtonema Hua plantlets could increase the synthesis of some concerned anzymes. The activity of protective anzyme system comprised of POD,SOD,CAT increased with the improvement of time and intensity of treatment.the activities of peroxidase(POD), superoxide dismutase(SOD) and catalase (CAT)increased to the highest level with all 15 hours, 17mTin each group. Then decreased with different degree. Because the protective anzyme system could eliminate the free radicals in plants, protect the nucleic acid and chromosome. So the increasement of its activity have great significant in the growth of plantlets.
