Sichuan snub-nosed monkey (Rhinopithecus roxellanae) is endemic to China . Because of the human's action such as deforesting , Snub-nosed monkey currently threatened by habitat loss, fragmentation . In order to devise efficiency conservation and management strategies for this endangered species , it is necessary to understand the genetic structure and molecular phylogeography . In this investigation , we studied parentage and genetic diversity in Sichuan snub-nosed monkey (Rhinopithecus roxellana) isolate population using microsatellite DNA markers , it was significant to protect and manage endangered species .In this investigation , 17 polymorphic microsatellite DNA primers identified in human were amplified in Sichuan snub-nosed monkey genomes, and found that 8 of these loci did not amplify product, 3 were monomorphic and 6 (D6S271, D6S287, D6S311, D11S1366, D17S1290, D6S493) were polymorphic . We compared 2 microsatellite loci sequences from human and monkey , it showed that these were same . Microsatellite DNA markers can use among species .In 84 samples , we found 36 alleles of 6 polymorphic microsatellite loci and the most allele number of D6S287 locus had 12 . We calculated the allele frequencies of 6 polymorphic microsatellite loci , and then we calculated the gene heterozygosity (H) and the polymorphism information content ( PIC ) and their mean of each locus . The values of gene heterozygosity is 0.523~0.838 , mean is 0.684 . The values of polymorphism information content is 0.348~0.71 , mean is 0.548 . That showed this isolate population had some genetic diversity and these loci were high polymorphism .We calculated the exclusionary power (first parent) is 0.101-0.354,total is 0.797684 ; Exclusionary power (second parent) is 0.174-0.533 total is 0.943441 . We did paternity testing of captive and wild isolate Sichuan snub-nosed monkey populations used 6 polymorphic microsatellite loci.In the study, we got the hair DNA genomes used the non-invasive DNA technology. Compared the amplfied result of hair DNA with the tissue DNA, we found they all had clear lines if hair number more than 3-5. These results implied the microsatellite DNA markers and non-invasive DNA technology can help establish the detailed pedigree and breed plan for isolate population of Sichuan snub-nosed monkey . And we could use this means to other endangered species for their conservation.In order to protect Sichuan snub-nosed monkey , perfect pedigree would help avoid the inbreeding and increase the genetic diversity.
