Fkbp11 was identified as a functionally novel gene through microarray analysis of hepatocellular carcinoma tissues from an HBx transgenic mouse model. It's a member of FK506 binding protein family, contains a conserved peptidyl-prolyl cis/trans isomerases. Our in vitro data suggested that Fkbp11 might involve in tumorigenesis by influencing the progress of cell cycle.In order to explore its in vivo function, we constructed a eukaryotic expression vector for Fkbp11 (IE-Fkbp11) based on pIRES2-EGFP, in which the expression of Fkbp11 is driven by a CMV promoter. Transgenic mice for Fkbp11 were generated by pronuclear injection of linearized IE-Fkbp11. Two founder mice were obtained for Fkbp11 after PCR and Southern blot analysis. Among them, one founder mouse P803 gave stable expression of Fkbp11 in pancrease, stomach, heart and kidney tissues, especially high level expression was found in acinar cells of pancrease, as demonstrated by Northern blot, western blot and fluorescence observation. Phenotype analysis of the P803 offsprings revealed abnormal changes in pancrease, stomach and kidney tissues. The expression of Fkbp11 in another founder mouse P823 was low and restricted in some acinar cells of pancrease, and its offsprings grow normally.In summary, we generated a transgenic mouse for Fkbp11 expression in multi-tissues, which provides us a model for studying the function of Fkbp11 in vivo.
