In nearly thirty years, micromanipulation technology had become a popular research hotpot, and microinjection is one of the most frequency-used micromanipulation. It was applied in Cell Engineering, Transgenic Engineering and modern biology and biomedicine widely. However, the microinjection is manipulated by men mostly so far, for its low automation. It is easy to get tired for the stuff, when they do the micromanipulation under the view of microscope for a long time. In this paper, we take the automation and intelligentization as the main theme, and build an automated micromanipulation system. Then the adherent cells are taken as the research subject to research microinjection methods.Firstly, it is very important to read a large amount of papers about the technology of microinjection and microinjection system, make a summary the advantages and disadvantages of research status review in oversea and domestic. Then analysis the mechanism of adherent cells microinjection and build a math model. Using the ABAQUS Finite Element Analysis Software to analysis the Stress, Strain, Deflection and Point-reaction force during cell microinjection.Then depends on the application and disadvantage of microinjection to build a hardware and software system.Next, make an introduction about platform system, then set up and calibrate the hardware system for the microinjection experiments. Beside, design the software framework and make a requirements analysis. In this paper, we use the VC6.0 as the development environment, combine with MFC library and Open CV library to package and module the hardware functions. Then an user interface was used to do the cell microinjection. In this program, some image process algorithms and other control algorithms was applied to control location platform, 3 DOF micromanipulator, microinjection pump,and so on, and all this methods are based on visual sever, such as pipette contact test and location, cell identification and location, and repeated location, and so on.Lastly, we use adherent fibroblast cells and osteoblast adherent cells as the experiment subjects. And a series of experiments would be carried out in this paper, such as pipette location experiment, cell injection volume tests, cells identification and counting experiment, cell microinjection experiment and cell repeated microinjection experiment, and so on. All these experiments results demonstrate that the cell microinjection methods in this paper improve the level of automation, accuracy and efficiency of cell microinjection. At last, the average cell injection accuracy achieves 80%, and the average cell injection rate is about 2-3 ones for each second.
