The White-headed langur is one of the endangered primate species in the world and first-class protected wild animal of China. In 2002 the White-headed langur is listed one of the 25 top priority protection primate species by World Animals Protection Organization. The population number of the white-headed langur is about 600 and the living condition is hard. By recent related study and report, this species has gained people's concern and interest. Hence, how to protect them has become very important in the future.The aim of article is to analyze the polymorphism of 5 micro satellite loci to obtain the genetic data of the White-headed langurs by extraction of high quality faecal DNA of White-headed langurs. Through analyzing these data we identify individuals and study the relationship of the White-headed langurs. So that the wild animal can be better studied and protected. During the research by efficient method of extracting faecal DNA we obtained high quality faecal DNA and successfully amplified micro satellite loci from the faecal DNA,and this extracting method reduce disturbance and hurt to the animals. At same time, we selected 5 pairs high polymophic micro satellite primer from 32 pairs primate micro satellite primer to amplify specific segment from the faecal DNA, and use SRY primers to judge sex of White-headed langur. Then we analyze the results of amplification by CERVUS2.0 software to make individual identification and paternity testing. The results show that: 1. In 72 samples, we found 45 alleles and the most allele number of D10s678 locus had 11. Based on allele frequency We calculated the gene heterozygosity(H) and the polymorphism information content (PIC ). the Mean value of H is 0.722; the Mean value of PIC is 0.687,which showed these loci were high polymorphism.2. The results showed 72 faecal samples of 5 groups White-headed langurs belonged to 46 different individuals; n group has 14 individuals; f group has 13 individuals; y group has 4 individuals, and all individuals are male;w group has 7 individuals;q group has 8 individuals.
