Study On Taxonomy And Identification Of The 26 Oceanic Actinomycate Strains Having Cytotoxic Activity

In this paper, we analysed genetic stability of cytotoxic activity of 26 Actinomycate strains which were fromer collected and had strong cytotoxic activity. The origin and the systematic evolution relationships of these strains were inverstigated.Most of the 26 Actinomycate strains which had strong cytotoxic activity had good genetic stability through generation transferring analysis, and the cytotoxic activity of strain 050642 and strain 060386 were strongest, with ID50 were 1270 and 1210,respectively. Through culture on media with sea water and distilled water, origins of the 26 strains were detected. Twenty five strains grew better in YE medium with sea water than with distilled water except for strain 0617230. So they were mostly likely to be marin origin. Based on morphological and physiological characteristics, strain 0617230 was proposed to genus Micromonospora and other 25 strains were proposed to genus Streptomyces.According to cell wall composition analysis,strain 0617230 belonged to amino acid typeâ…¡and Sugar type D, other 25 strains belonged to amino acid typeâ… and Sugar type C.According to phosphelipid types analysis of 8 strains, all of them had PE only, so they belonged to phospholipoid typeâ…¡.Through 16S rDNA sequence analysis of the 26 strains, strain 060386 was the most similar of 99 % to Streptomyces griseoincarnatus and Streptomyces griseoflavus; strain 050643 was the most similar of 96 % to Streptomyces violaceolatus; strain 060524 was the most similar of 98 % to Streptomyces glomeratus; strain 060519 was the most similar of 97 % to Streptomyces fungicidicus,Streptomyces sampsonii,Streptomyces albidoflavus; strain 061342 was the most similar of 98 % to Streptomyces sp. 28a-5-1 that had not be confirmed to be which species was; strain 061303 and other 11 strains were the most similar of 98 % to Streptomyces sp. 28a-5-1 that also had not be confirmed to be which species was; strain 050642 and other 9 strains were the most similar of 98 % to Streptomyces cheonanensis; however, strain 0617230 was proposed to genus of Micromonospora with the most similar of 97 % to Micromonospora echinospora.In order to know systematic evolution relationships of these 26 Actinomycate strains, we used polyphasic taxonomy method.The rusluts showed consistency of morphological,physiological and cell wall chemotaxonology characteristic, with 16S rDNA sequences.
