objective: We construct an eukaryotic expression recombinant containing the rat IL-6 gene to regulate IL-6 gene expression with RNAi technique. Methods: The rat IL-6 cDNA was reverse transcripted from total RNA which was extracted from rat's colon, amplified by PCR, then it was inserted into the vector pcDNA3.1/CT-GFP-TOPO and transfected into COS-7 cells.Result: Sequencing results show that an eukaryotic expression vector, pcDNA 3.1/CT-GFP-TOPO/IL-6 was correctly constructed. After transfection, we can see green fluorescent in COS-7 cells through fluorescence microscope. Conclusion: Successful construction of rat IL-6 eukaryotic expression vector creates an opportunity for us to dissect the relationship between IL-6 and ulcerative colitis by RNAi technique.
