Interleukin-2 (IL-2) is one of the polypeptide produced by T cells in immune system, which has 155 amino acids. IL-2 acts as a modulator in immune system, especially as a growth factor modulating proliferation and differentiation of lymphocyte cells. It can influences livability of the neurons and neuroglias. IL-2 (5 nmol.L-1) increased proliferation rate of oligodentrocytes and it's glia cell line for 2.5 times. IL-2 has nutrition function on the neurons. Awatsuji et al cultured neurons of different part of the brain of embryonic rat in vitro, and the results showed IL-2 (200 U.mL-1) affiliated proliferation of hippocampi cells, prolonging the living time of neurons. The research results of our lab suggested that IL-2 affect proliferation of anterior pituitary cells and pituitary tumor cell lines, which had a relationship with sex hormones. Recombinant IL-2 is used as a clinical immunepreparation to several kinds of tumors. It can affiliate the basal secretion of ACTH, PRL, TSH in tissue pieces of pituitary, having a function on the secretion of LH, FSH, GH simultaneously.Estrogen (E2) is one of the steroid hormones, which is mainly synthesized and secreted by the gonad affecting proliferation, differentiation and function exerting of target tissues. Circumferential estrogens were mainly produced by ovary and spermary, and the brain estrogens came mostly from hypothalamus, neurons of limbic system and neuroglias. There are a and 3 subunits of estrogen receptor(ER) subtypes. They are different in their distribution, density and mediated action. ER can be expressed in nucleus or in cellular membrane. In addition to the classical genomic pathway through nucleus receptors, estrogen can cause to produce some cellular rapid membrane-mediated effects through nongenomic pathway.HeLa is a cervical carcinoma cell line established by Gey from a 31 years old Negro woman in 1951, which is an epidermis-like cell line. The incidence of a cervical carcinoma is just smaller than breast carcinoma in the world malignancies of women. The women caught a cervical carcinoma aging from 15 to 85, and mainly above 45 years old. These years, young catchers (less than 35) increased significantly. Bcap-37 is a breast cancer cell line that first established in China.The experiment purpose is to confirm the influence of IL-2, E2 and the agonist/antagonist of E2 tamoxifen (TAM) on proliferation of cervical cells of rat, HeLa cervical carcinoma cell line and Bcap-37 breast carcinoma cell line as well as the possible mechanism of therefunction.The following methods are adopted in the experiment1. The primary culture of cervical cells of rat.2. The culture of HeLa cervical carcinoma cell line and Bcap-37 breast carcinoma cell line.3. Drawing growth curves through cell counting.4. We used immunocytochemistry to explore the expression of IL-2R.5. All cells were cultured for enough days to get the total RNA. The cDNA was synthesized from the total RNA, and the PCR was used. Through this method, IL-2RmRNA and ERmRNA were detected.6. The DNA sequence was assayed to determine whether the sequence of ER ? was we wanted.7. We adopted MTT assay to observe the effects of IL-2, E2 and TAM on the OD value of the cells.8. Flow cytometry was used to determine the influence of IL-2, E2 and TAM on the DNA content of different stages of cell cycle.9. And laser scanning confocal microscope was also adopted to investigate the influence of IL-2, E2 and TAM on level of the intracellular Ca2+.The results are as follows:1. IL-2(100 U.mL-1) and E2(1X10-6 mol.L-1) shifted the growth curves of HeLa cervical carcinoma cell line and Bcap-37 breast carcinoma cell line upward. But TAM (10-7 mol.L-1) shifted the curve of HeLa upward, and shifted the curve of Bcap-37downward.2.
