[Objective] To study on the effects of the recombinant eukaryotic expression plasmid pcDNA3.1(+)-ChIL-15to the immunological function in chickens was to lay the foundation for clinical application of the recombinant ChIL-15.[Methods] According to ChIL-15mRNA sequence published in GenBank, a pair of specific primers was designed, the total RNA which was extracted from chicken spleen was treated as templates and the ChIL-15gene was amplified by RT-PCR. The pcDNA3.1(+)-ChIL-15was Constructed with pcDNA3.1(+). The effects of the recombinant eukaryotic expression plasmid pcDNA3.1(+)-ChIL-15to the immunity index of the chicken were detected periodically.[Results] The ChIL-15gene fragment was581bp.The results show the amplification of ChIL-15gene was successful. The results of the double digest and the PCR identification shows the recombinant eukaryotic expression plasmid pcDNA3.1(+)-ChIL-15was successfully constructed. The results of the Animal test show pcDNA3.1(+)-ChIL-15could enhance the active lymphocyte cells percentage in the peripheral blood, and clearly enhance NDV, IBD and NIBV vaccines to induct the antibodys, increase spleen index, and protect chickens from NIBV.[Conclusion] Eukaryotic expression plasmid pcDNA3.1(+)-ChIL-15could increase the immunity of chicken, and enhance the immunity of NIB, IBD and ND vaccines.
