Nucleic acids are the basic ingredient of all the biologic cells and they have an important function in life processes, such as growth, propagation, descendiblity and contabescence. Study of chemical components, structure and biologic function of nucleic acids is very important and quantitative determination of nucleic acids is the base of investigation on nucleic acids. Nucleic acid analysis continues to be an important and active area of investigation in the fields of development of medicines, study of biochemistry reaction, diagnosing, treating and preventing disease.In this paper, the progress of analytical technique and quantitative determination of nucleic acid has been reported in detail, as well as the binding mechanism of nucleic acid and micromolecule by fluorescent spectrometry. On the basis of the dyes as a spectral probe of nucleic acid determination, we have studied the analytical chemistry performance of the trace nucleic acids using spectrophotometry, resonance light scattering and fluorescence method. Those methods have been successfully applied to the nucleic acids determination in samples. At the same time, their mechanisms are also discussed by fluorescence method, including binding constant, binding mode, sorts of fluorescence quenching and the main sorts of binding force. Some novel achievements are obtained.This paper features in systematic, scientific and creative methods.1. Systematic featureIn this paper, the reaction conditions (such as acidity, medium, amount of developer, influence of organic micromolecules, amount of dyes, et al) of spectral probe with nucleic acids by spectrophotometric, resonance light scattering and fluorescence methods have been researched systematically. Based on this, a series of methods of determination of nucleic acids were established and all the methods have good selectivity and high sensitivity.2. Scientific featureThe method mentioned in the paper has been successfully applied to the determination of nucleic acids in synthetic samples and real samples. The results were satisfactory.
