Resonance Light Scattering (RLS) is a novel analytical method of the molecular emission spectrometry field. For its remarkable characteristics of high sensitivity, simple operation and good selectivity, RLS brings to more attention and interesting. In recent years, RLS has been studied and applied in the fields of biology, environmental science, and material science and so on. Especially in the field of studying biological macromolecules, RLS becomes an important new approach following absorption spectrophotometry and fluorescence. Now organic dyestuff is most regarded as the probe of light scattering. In this thesis, we studied the interaction between bovine serum albumin (BSA) and the probe of organic dye (aurintricarboxylic acid), compound complex (Chrome azurol S-Fe(III)) and organic acid (sulfosalicylic acid) by RLS. In fact, RLS technique is the same as that of resonance fluorescence in experiment, so we studied the difference and relationship between RLS and resonance fluorescence of Butyl Rhodamine B (BRB).The main contents of the thesis are as follows:1. RLS characteristics, the influencing factors and the optimum conditions of the interaction between Aurintricarboxylic Acid (ATA) and Bovine Serum Albumin (BSA) were studied. The RLS intensity of ATA is very weak, but the RLS signal is enhanced when trace BSA was added into the system, the maximum scattering peak lies at 341nm. The intensity of RLS in the system is direct proportion to the concentration of BSA. In optimal conditions, the determination limit of BSA is 20ng/ml, linear range is 0~0.6μg/mL. The satisfactory results have been achieved with this method in the quantitative analysis of proteins in synthetic sample and human urine sample.2. RLS characteristics of the interaction between Chrome Azurol S (CAS)-Fe (III) and Bovine Serum Albumin (BSA) were studied. Based on this, a method of determination of trace BSA was established. The influencing factors and the optimum conditions of the system were also studied. The intensity of RLS at 340 nm is directly proportional to the Concentration of BSA in the range 0~4.0μg/mL, the determination limit is 20 ng/mL. The method has applied to determination of BSA in synthetic samples with satisfactory results.3.
