This study attempted to find out the effects of sfrp3 overexpression on mouse early tooth development,then, we expected to more understand the role of Wnt signaling in the process of tooth germ induced. Wnt family have many members ,their Antagonists and their receptors in tooth germ.All explained that Wnt signaling may be very important during origination of tooth development. sFRPs were found proteins recently.In function, sFRPs may control Wnt signaling. Different sFRPs recognises and combines with special Wnt to control Wnt signaling.In our prophase research,we have found that sfrp3 and Wnt5a were expressed in molar. Get function and lose function are often used of Studying the function of gene.The methods include gene knock out, RNA interfere, gene Overexpression and so on.We Overexpressed sfrp3 through lentivirus vector(HIV-1),and sFrp3 would break transmitting of Wnt signaling.In the research, the studies included pNL -IRES2-EGFP-sfrp3 was constructed, Lentivirus was producted, and sfrp3 was Overexpressed in molar mesenchyme with lentivirus as vector.The results demonstrated that lentivirus infected 56% tooth germ mesenchyme cells, and they may Overexpress sfrp3 with EGFP as report gene;At E13.5, Overexpressing sfrp3 in molar mesenchyme with lentivirus as vector had no distinc effect for tooth development.
