The tooth development is dependent on the mutual induction between dental epithelium and mesenchymal. Various signaling pathways, in particular, Wnt signaling pathway plays an important role in the process of mutual induction.However,the research about the specific role and mechanism of the Wnt signaling pathway in tooth development remains to be improved.In this essay,we discuss the role of Wnt signaling pathway antagonists sFrp2 and sFrp3 in the development of mouse molar by applying lentivirus-mediated overexpression in dental mesenchymal cells. Real-time PCR results showed that the expression level of sFrp2 and sFrp3 were increased by 270% and 220%, Axin2 expression levels of about 60% reduced.The results above indicated that the efficiency of overexpression of sFrp2 and sFrp3 was good, and the classical Wnt signaling pathway was inhibited. When analysising the transplanted tissue of different days, We discover that the process of mouse molar development was accelerated, the process of ameloblast differentiation and enamel matrix secretion was promoted, the thickness of enamel and the size of the final form of tooth were increased,while the expression of sFrp2 and sFrp3 was accelerated. It was found that, sFrp2 and sFrp3 while over-expression makes the expression of Shh and Bmp4 were reduced by 62% and 52%, indicating that the inhibition of classic Wnt signaling pathway may affect the expression of downstream genes Shh and Bmp4. This experiment proved that, Wnt signaling pathway plays an important role in mouse molar development, especially the development of ameloblast.
