The white-headed leaf monkey is a rare primate species of on the verge of extinction in the world, which lives only in the karst stone areas within Chongzuo City in the south of Guangxi. In 2002 the white-headed leaf monkey is listed one of the 25 top priority protection primate species by World Animals Protection Organization. In order to scientifically and effectively conserve this species, analyzing its genetic diversity is necessary. But up to now, the genetic diversity of the white-headed leaf monkey is not reported in China and in the World. The396bp hypervariable control region sequence in the left of the mitochondrial(mt)DNA genome from 31 samples(there are at least 9 individuals from 9 population) of white-headed leaf monkey were collected from Chongzuo Banli nature reserve, Fushui Bapen nature reserve of Chongzuo city. The 9 different individuals of the 31 sample were sequenced and population analysis was conducted to assess the level of genetic diversity of present . The results showed: (1)In this study, the length of the D-loop of mtDNA genome from 9 samples was 396bp.15 site were polymorphic. Among them, The site of transition and transversion were 13 and 1 respectively. There was 1 site with three base and nucleotide diversity was0.01167. (2)7 haplotypes were identified from 9 sequences.(3) Among the population of three geographical unit of the White-headed leaf monkey, the JCHSH geographic region have higher numbers of base differences, most high of nucleotide diversity and most high of average number of nucliotide differences. Based on these results, we suggest some coservation and management measurements must be taken to effect (1) In Jiuchongshan region, the population of the white-headed leaf monkey was relatively larger. And genetic diversity was abundant. Therefore, In term of protecting gene diversity of this population. We propose classifying it as the unit of high priority protection. (2) the genetic diversity of Buzun and Longguanshan was lower than Jiuchongshan's. We propose and then the order of protection of these population are protected.
