Malignant tumor is one of the most deadly diseases for human. Scientists from both home and abroad having been done many researches to conquer it, among the theories and methods, ultrasound especially focus ultrasound is the most promising way. Since Japanese Umemura put forword Sonodynamic Therapy ( SDT ) for the first time, SDT had developed greatly for its potential of clinic application . Ultrasound especially focus ultrasound can penetrate deeply in organic tissues without any harm; Hematoporphyrin Derivatives ( HpD ) have priority concentration to tumor tissues and maintain for a long time; Meanwhile, ultrasound can activate HpD and produce some substance such as '62 which has oxidative activation to damage even kill tumor cells.In order to cure tumor in human's body and promote SDT's application to clinic, this paper designed to sift out the optimum ultrasonic frequency, the optimum radiation time and the optimum suspension in which SW-480 cells exposed to ultrasound, the other goal is to identify whether or not SDT can cause cancer cells apoptosis accompany with necrosis, whether or not lipid peroxidation appears during the course of SDT, and whether or not the reproduce potential of cancer cells drops correspondingly.We have used MTT to study the survival of SW-480 cancer cells after expos to ultrasound, and MTT had been proved to be a good way to measure the activity of c ils. The author has done research on cultured in vitro SW-480 cancer cells under the certain focus ultrasonic intensity( 1.5W/cm ), and the experimental results show: (l)When the ultrasonic frequency was 1.6MHz, the minimum survival rate of SW-480 cancer cells in RPMI1640 medium after 120 seconds' radiation was 51.06% compared with control cells; (2)When exposed to 1.1 MHz focus ultrasound for 120 seconds, we can observe compensatory rising respiration after 16 hours' culture.Based on this, the article adopted H-E staining, PI-HO dual fluorescent staining and scanning electron microscope to observe the morphologic changes of the cancer cells after being radiated by ultrasound, together with TEA and clone forming test, we observed significant apoptosis phenomena accompany with necrosis. In addition, we represented that there was lipid peroxidation during the course of ultrasound activating HpD, and the reproduce potential of cancer cells dropped greatly. Those parameters should be very important for further research on both in vitro and in vivo experiments as well as provide a new evaluation index for Sonodynamic Therapy.
