The sporocarp formation of Asterophora lycoperdoides and purification and isolation of a-amanitin were studied in this research. In the first part, it was first observed that the sporocarps could be formed from cultivated hyphae of A. lycoperdoides in our laboratory, with dispersive arthrospores and catenular chlamydospores. The HSVA was identified to be the optimized culture medium by the experiment of filtrating basic culture medium. And the optimized cultivating conditions were glucose for carbon, NH4CI, (NH4) 2HPO4, NH4NO3 for nitrogen, temperature in 20-25C, and pH in 6-9, by the single factor experiment of the HSVA. According to our orthogonal experiment, the optimized culture medium was developed from HSVA, and the variable factors were 3.0 g/L glucose, 0.2g/L NH4CI, pH 5.0, and the growing temperature at 20C. The result showed that A. lycoperdoides was not obligatory parasitism as reported before. Instead, it could grow naturally in semi-synthetic medium. Thus, it could be used as the material of the research of sporocarp formation of mushrooms, because of its simple cultivating conditions and easily being controlled in laboratory. Furthermore, its water extract showed an inhibitory effect on the budding of Brassica napus and Phaseolus aureus, with the efficiency of 71.27 % and 56.33 %, respectively. This effect may potentially be applied on the development of new biological growth regulator.In the second part, the Amanita virosa was gathered in the place where poisoning of the people were found, and the research of morphology and taxology ofit were studied. Furthermore, the toxicity experiment of rats showed the lethiferous toxin was contained in it, and later the a-amanitin was identified from it. The united equation that Y = 5159X + 1097. 4 was built, by the establishment of two standard curves of the gradient mark of the a-amanitin and the gradient mark of the a-amanitin added with liquid from A. virosa, and followed making the significance tests of them to identify them were from one group. The content of a-amanitin of the A. virosa is 2833.8ug/g (dried sporocarp) by mensurating it. The results showed that no a-amanitin had been detected from the sporocarp of A. lycoperdoides and A. nivalis by RP-HPLC. But in the sporocarp of Amanita excelsa, a-amanitin could be found, and its content was 74.0ug/g (dried aporocarp).
