This research studied the differentiation culture of the Eustoma russellianumlaminae via different concentration and combination of the incretion influencing thewhite Eustoma russellianum laminae. Then we can establish the whole system of theheredity transformation and provide a good foundation for the next step. At thesame time, the examination established the most proper choosing concentration andpre-cultured time of transforming the laminae via studying the sensitivity of thelaminae to Kanamycin, the different pre-cultured time influencing differentiation andthe influence of reducing the sensitivity of the antibiotic. The examination clonedthe special promoter P CHSA and directionally inserted the plasmid pBI121 (whichobtained Lc) which replace the 35s promoter in the original plasmid. Theexamination created a new carrier pBI121-1 which used in the research of theflowers'color. In the base of the earlier stage,we transformed Eustoma russellianumwith pBI121 and pBI121-1 separately. And then we got the transformation offspringfor analyzing them with molecular biology. Finally, we can filtrate and get thetransgene offspring.The main results of the examination:1. The best differentiation culture of the white Eustoma russellianum laminae is MS+6BA1mg/L. We found the laminae are very sensitivity to NAA which can't differentiate in the culture contained NAA 0.01mg/L.2. We used the total DNA of Petunia hybrida as the stencil and cloned the special promoter P CHSA. We directionally inserted it into the plasmid carrier pBI121 (the aim gene is Lc)to replace the original CaMV 35s promoter and created the expression carrier contained lc gene, which can ameliorate flowers'color is pBI121-1.3.
