. Promoter is a DNA sequence used for RNA polymerase combinating , and is an important cis-acting element in the regulation of the gene expression. It locate at upstream of initiation site of transcription. The rd29A gene is a stress-inducible gene isolated from a model plant- Arabidopsis thaliana, and the promoter is an inducible promoter. The promoter of the rd29A gene is amplified and from Arabidopsis thaliana genome by polymerase chain reaction and cloned into pUC19 vector. Sequence analysis showed that the clone of promoter fragment contained 937 nucleotides and shared a sequence homology of 99. 8% with the reported rd29A promoter.
