The different organs of Elaeagnus mollis Diels were used in this experiment as the explants. After establishing the aseptic propagation system, the aseptic tube stems and leaves were used to study the effect of different exogenous hormones combination and different culture conditions on organgenesis. The optimum medium and culture condition was screened for fast propagation of Elaeagnus mollis Diels. In this experiment, some conclusions were drawn. After being steriled, explants were treated by using antibiotic substance solution with a level of l00mg/L for 30 min, the non-contamination rate was up to 80%-92%. The effect was appropriate. The suitable medium of Elaeagnus mollis Diels was MS+BA0.5+NAA0.01 through tube stem to induce adventitious buds. The optimum medium of Elaeagnus mollis Diels was MS+BA1+2.4-D1.5 through tube leaves to induce callus. The optimum medium of Elaeagnus mollis Diels was MS+BA1+MBA0.05 through callus to induce buds differentiation. The suitable medium of plantlet rejuvenation was MS+BA0..5+IBA0.4. The treatment of single factor of IBA1.5 showed a better effect on striking root of tube shoots. In inducing callus process, the suitable illumination conditions were the former period with darkness culture and the later period with light culture. In inducing dedifferentiation and redifferentiation process, the temperature of 24-26℃ was suitable. In the stage of striking root, the optimum temperature was 25-28℃ and the intensity of illumination was 1000-20001x with 16h. This experiment also studied the transplanting technology of Elaeagnus mollis Diels tube plantlet and environmental conditions and the transplanting survival ratio was up to 66%.
