Part 1: The most important task of the post-genomic era is to investigate the expressed products of geneme. Proteome indicates the PROTEins expressed by a genOME or tissue.The technology of proteomics requires the seperation, indentification of large number of proteins. Currently the most powerful method to separate proteins is 2D-electrophoresis. Thus 2D PAGE is the heart of proteome technology. In proteome projects, which aim to identify and characterize all proteins expressed by an organism or tissue, the identification of proteins is central. Indentificatiort is also the first step towards studies on protein co- and post-translational modification, and ultimately, function.In the present study, the total proteins of the photo-thermo sensitive genie male-sterile rice (ORYZA SATIVA, peiai64S) spikelet at meiosis stage were used as the material. By optimizing crucial factors and procedures such as sample treatment, electrophoresis parameters,and gel concentration, 2-D maps with high quality and reproducibility were obtained. Mean standard deviations of about 1. 49+0. 22mm for IPG-IEF and 1. 24 ?0.18mm for SDS-PAGE were obtained. 57 protein spots out of about 1000 detectable spots on the 2-D gels were indentified by the following two methods:l)N~terminal Edman degradation microsequencing after the protein spots were electro-transferred to PVDF membrane. 2)MALDI-TOF-MS peptide fingerprint analysis of the protein spots and protein database searching. The 2D protein maps of the rice spikelet during the sterile and fertile stages were compared. While the rice spikelet changed from sterile to fertile stages, about 16 protein spots showed variations that were significant and reproducible. Six up-refulated proteins and nine down-regulated proteins were identified. One protein only exist in fertile stage.
